GD has developed a PRRSv challenge model using a field strain isolated from a pig farm in the southern part of the Netherlands in 2022. The strain was initially isolated on PAM cells and further propagated on MARC145 cells. Additionally, we performed whole-genome sequencing and developed a virus neutralization test for this strain.

The animal model was developed in 7-week-old piglets. Upon nasal inoculation, an increase in rectal temperature was observed, along with mild respiratory clinical signs. Viraemia, measured by strain-specific qPCR, was observed in serum of all challenged piglets up to 21 days post infection. Furthermore, all lungs of infected piglets were positive by qPCR at 10 days post-infection, and 70% of the lungs at 28 days post-infection.
